HPLC法同时测定委陵菜中5个黄酮类成分的含量

目的:建立HPLC 法同时测定委陵菜中芦丁、金丝桃苷、槲皮素、芹菜素和山柰酚5 个成分含量。方法:采用ZORBAX SB-C18(4.6 mm×250 mm,5 μm)色谱柱,以乙腈-0.2% 磷酸水溶液(5:95)为流动相A,乙腈-0.2% 磷酸水溶液(70:30)为流动相B,梯度洗脱,流速为1 mL·min-1,检测波长为320 nm。结果:在上述条件下,芦丁、金丝桃苷、槲皮素、芹菜素和山柰酚的质量浓度分别在12.50~200、6.25~100、6.25~100、6.25~100 和4.00~64.00 μg·mL-1 范围内与峰面积呈良好的线性关系,平均加样回收率(n=9)均高于95%,RSD 小于3.0%,10 批样品中芦丁、金丝桃苷、槲皮素、芹菜素和山柰酚含量范围分别为144.7~342.4、117.9~232.7、123.3~203.6、65.7~142.6、43.3~89.3 μg·g-1结论:方法学验证结果表明,本法适用于委陵菜中黄酮类成分的含量测定。

Objective:To establish a method for simultaneous determination of rutin,hyperin,quercetin,apigenin and kaempferol in Potentilla chinensis Ser.by HPLC.Methods:HPLC separation was carried on a ZORBAX SBC 18 column(4.6 mm×250 mm,5 μm)with the mobile phase consisted of mobile phase A[acetonitrile-0.2% phosphoric acid solution system(5:95)] and mobile phase B [acetonitrile-0.2% phosphoric acid solution system (70:30)] in a programme of gradient elution.The flow rate was 1 mL·min-1 and the detection wavelength was set at 320 nm.Results:Under the above conditions,the linear ranges of rutin,hyperin,quercetin,apigenin and kaempferol were 12.50-200 μg·mL-1,6.25-100 μg·mL-1,6.25-100 μg·mL-1,6.25-100 μg·mL-1,and 4.00-64.00 μg·mL-1; the average recoveries(n=9)for the sample preparation of the markers were more than 95.0% and RSDs were less than 3.0%;the content ranges of rutin,hyperin,quercetin,apigenin and kaempferol in 10 batches of samples were 144.7-342.4,117.9-232.7,123.3-203.6,65.7-142.6 and 43.3-89.3 μg·g-1,respectively.Conclusion:The established method is proved by methodology validation to be suitable for content determination of flavonoids in Potentilla chinensis Ser.
