芳香新塔花和小新塔花全草中4个成分含量测定及聚类分析

目的:建立芳香新塔花和小新塔花中薄荷酮、异薄荷酮、薄荷醇和胡薄荷酮的含量测定方法,比较不同种、不同产地的新塔花中4个成分的含量差异。方法:采用气相色谱法,HP-FFAP毛细管柱(30 m×0.32 mm×0.25μm),载气N2,流速25 mL·min-1,程序升温(初始温度70℃,保持1 min,以8℃·min-1的速率升温至160℃,保持1 min),进样口温度250℃,检测器温度250℃,测定4个成分的含量,应用DPS 2000软件对实验数据进行聚类分析。结果:芳香新塔花中4个成分含量:胡薄荷酮>异薄荷酮>薄荷酮>薄荷醇;小新塔花中4个成分含量:胡薄荷酮>异薄荷酮>薄荷醇>薄荷酮;芳香新塔花中4个成分的含量高于小新塔花;应用聚类分析法,可以将芳香新塔花和小新塔花分开。结论:本方法操作简单,准确,可用于不同种及不同产地的新塔花药材中成分的含量测定。

Objective:To establish a method for determination of menthone,isomenthone,menthol,and pulegone of Ziziphora clinopodioides and Z.tenuior,and compare the content difference of the four components in Ziziphora of different habitats and different species.Methods:In order to determine the content of the four components,the gas chromatography and the cluster analysis by DPS 2000 software were adopted.An HP-FFAP capillary column (30 m×0.32 mm×0.25 μm) was used,and the carrier gas was N2 with the flow rate of 25 mL·min-1.Column temperature was raised by programming,(initial temperature at 70℃,held for 1 min;raising at 8℃·min-1 to 160℃ held for 1 min),the injector temperature was at 250℃ and the detector temperature was at 250℃.Results:The contents of 4 components in Ziziphora clinopodioides were pulegone> isomenthone> menthone> menthol.The contents of 4 components in Z.tenuior were pulegone> isomenthone> menthol> menthone.And the contents of 4 components of Ziziphora clinopodioides were higher than those of Z.tenuior,Z.clinopodioides and Z.tenuior could be separated by cluster analysis.Conclusion:This method is simple and accurate,and can be used for determining the components in Z.clinopodioides and Z.tenuior of different species and different habitats.
