斯皮诺素表观油水分配系数的测定

目的:测定斯皮诺素在正辛醇-水系统中的表观油水分配系数,系统考察pH对表观油水分配系数的影响。方法:采用HPLC法测定斯皮诺素的含量,并采用摇瓶法测定斯皮诺素在不同pH的正辛醇-水中的油水分配系数。结果:在37 ℃下,斯皮诺素在pH 为1.3、3.3、4.0、5.0、6.8、7.0、7.4、8.0 时,表观油水分配系数平均值分别为2.34、2.54、2.66、2.67、2.69、2.80、3.62、5.03。结论:斯皮诺素在不同pH条件下油水分配系数略有区别,但logP值小于1,油水分配系数过小可能是造成斯皮诺素在胃肠道全段通透性较差的主要原因。

Objective: To determine the apparent oil-water partition coefficient of spinosin in octanol-water system and systematically investigate the influence of pH on the apparent oil-water partition coefficient. Methods: The content of spinosin was determined by HPLC and the apparent oil-water partition coefficients for spinosin in octanol-water system at different pH values were determined using the shaking flask method. Results: When the pH value of the solution was 1.3,3.3,4.0,5.0,6.8,7.0,7.4,8.0 at 37 ℃,the corresponding average value of the partition coefficient of spinosin was 2.34,2.54,2.66,2.67,2.69,2.80,3.62,and 5.03,respectively. Conclusion: The partition coefficient of spinosin varied little in different pH conditions. However,the logP was consistently less than 1,and the fact that the partition coefficient was rather small may be the main reason for the poor intact gastrointestinal permeability of spinosin.
