目的: 建立离子色谱法测定羧甲基壳聚糖中有害物质二甘醇酸。 方法: 采用IonPac AS 9-HC色谱柱(250 mm×4 mm),以12 mmoL·L-1碳酸钠溶液作为淋洗液,等度洗脱,流速为1.0 mL·min-1,抑制电导检测器。 结果: 二甘醇酸在2.05~51.2 μg·mL-1范围内线性关系良好(r=0.9999);平均回收率(n=6)为98.8%,RSD为1.2%。医用羧甲基壳聚糖原料中含有一定量的二甘醇酸。 结论: 本方法准确,灵敏度高,重复性好,无污染,可用于医用羧甲基壳聚糖原料的质量控制。

Objective: To develop an IC method for the determination of diglycolic acid(DGA),a harmful substance in carboxymethylated chitosan(CMC). Methods: An IonPac AS 9-HC column(250 mm×4 mm) was used.The eluant was 12 mmol·L-1 sodium carbonate and the flow rate was 1.0 mL·min-1.The detector was conductivity detector. Results: DGA showed good linearity in the range of 2.05-51.2 μg·mL-1(r=0.9999).The average recovery(n=6) was 98.8%(RSD=1.2%),respectively.Both the materials of CMC for medical use contained a certain amount of DGA. Conclusion: The method is accurate,sensible and reproducible.The developed method can be used for the quality control of CMC.
