目的: 建立同时测定银黄含片中6个咖啡酰奎宁酸类成分的HPLC方法。 方法: 采用Inertsil ODS(4.6 mm×250 mm,5 μm)色谱柱,以乙腈(A)-0.4%磷酸溶液(B)为流动相,梯度洗脱(0~15 min,5%A→20%A;15~30 min,20%A→30%A;30~60 min,30%A),流速1.0 mL·min-1,检测波长为327 nm。 结果: 3-O-咖啡酰奎宁酸、绿原酸(5-O-咖啡酰奎宁酸)、4-O-咖啡酰奎宁酸、4,5-O-二咖啡酰奎宁酸、3,5-O-二咖啡酰奎宁酸、3,4-O-二咖啡酰奎宁酸进样量分别在0.04~0.69,0.04~0.67,0.04~0.68,0.02~0.33,0.01~0.15,0.04~0.70 μg范围内与峰面积呈良好的线性关系;平均回收率(n=6)分别为95.0%,99.1%,98.9%,98.9%,96.2%,103.5%。 结论: 本法操作简便,结果准确,灵敏度高,可用于银黄含片中多指标成分的质量控制。

Objective: To develop a quantitative method for simultaneously determining six caffeoylquinic acids(CQAs) in Yinhuang buccal tablets by HPLC. Methods: Inertsil ODS(4.6 mm×250 mm, 5 μm) column was adopted.The mobile phase consisted of acetonitrile(A)-0.4% phosphoric acid solution(B) with gradient elution(0-15 min,5%A→20%A;15-30 min,20%A→30%A;30-60 min,30%A) at the flow rate of 1.0 mL·min-1. The detection wavelength was 327nm. Results: The calibration curves were linear in the ranges of 0.04-0.69,0.04-0.67,0.04-0.68,0.02-0.33,0.01-0.15,0.04-0.70 μg for 3-O-caffeoylquinic acid(3-O-CQA),chlorogenic acid(5-O-caffeoylquinic acid,5-O-CQA),4-O-caffeoylquinic acid(4-O-CQA),4,5-O-dicaffeoylquinic acid(4,5-O-diCQA), 3,5-O-dicaffeoylquinic acid(3,5-O-diCQA),3,4-O-dicaffeoylquinic acid(3,4-O-diCQA); The average recoveries(n=6) were 95.0%, 99.1%, 98.9%, 98.9%, 96.2% and 103.5%, respectively. Conclusion: The method is simple, accurate, sensitive and can be used for quality control of multicomponent in Yinhuang buccal tablets.
