高效液相色谱法同时测定北败酱草中6个成分的含量

目的: 建立同时测定北败酱草中绿原酸、秦皮乙素、咖啡酸、木犀草苷、木犀草素和芹菜素6个成分含量的方法,为制定质量标准提供依据。方法: 采用高效液相色谱-二极管阵列检测法,使用Waters Symmetry C18色谱柱(250 mm×4.6 mm,5 μm),以甲醇-0.4%磷酸水溶液为流动相,进行梯度洗脱,流速1.0 mL·min-1,柱温35℃,检测波长340 nm。结果: 采用所建立的方法,6个成分分离度良好,平均加样回收率为96.8%~104.9%,RSD为0.6%~2.6%,符合定量分析要求。10批样品中绿原酸、秦皮乙素、咖啡酸、木犀草苷、木犀草素和芹菜素6个成分含量范围分别为148.0~3 185.4、7.9~17.9、5.0~175.1、16.3~1 411.5、24.3~2 367.7、5.6~74.8 μg·g-1,含量差异较大,其中绿原酸、木犀草素和木犀草苷可作为北败酱草质量控制的指标性成分。结论: 建立的分析方法适用于北败酱草的质量控制与评价。

Objective: To establish a method for simultaneous determination of six components of chlorogenic acid,quercetin,caffeic acid,luteoloside,luteolin and apigenin in Sonhi Arvensis Herba,and to provide a basis for the establishment of quality standards. Methods: High performance liquid chromatography-photodiode array (HPLC-PDA) method was used. The separation was performed on Waters Symmetry C18 column(250 mm×4.6 mm,5 μm). The mobile phase consisted of methanol-0.4% phosphoric acid solution with gradient elution at a flow rate of 1.0 mL·min-1. The detection wavelength was 340 nm,and the column temperature was 35℃. Results: The six components were well separated,the average recovery rates were 96.8%-104.9% with RSDs of 0.6%-2.6%,which met the requirements of quantitative analysis. The content ranges of chlorogenic acid,quercetin,caffeic acid,luteoloside,luteolin and apigenin in 10 samples were 148.0-3185.4 μg·g-1,7.9-17.9 μg·g-1,5.0-175.1 μg·g-1,16.3-1411.5 μg·g-1,24.3-2367.7 μg·g-1 and 5.6-74.8 μg·g-1,respectively. The contents of the six components in 10 samples varied greatly,and chlorogenic acid,luteolin and luteoloside could be used as the index component to evaluate the quality of Sonhi Arvensis Herba. Conclusion: The established analytical method is suitable for the quality control and evaluation of Sonhi Arvensis Herba.
